The museum was inaugurated on August 30, 1926, amid a broader Argentine movement during the 1910s and 1920s to create provincial museums. It was founded by Pedro E. Martinez, a prominent local lawyer, professor, and philanthropist, who served as its first director until his death in 1935. In 1938, the Province officially named the institution Dr. Pedro E. Martinez to honor his foundational work. Originally housed at the Centennial School of Parana, the museum occupied several transient locations before moving to its permanent site in 1980. In 2012, a plaque was placed on the premises identifying the building as a Site of Memory, acknowledging its dark history as a clandestine detention center during the Argentine dictatorship between 1978 and 1980. The museum celebrated its centenary in 2026, marking the occasion with the inauguration of the heritage exhibition Un destello sostenido.

The museum safeguards a heritage collection of more than 1,200 works of art spanning multiple disciplines, including painting, drawing, printmaking, sculpture, ceramic art, and textile art. The holdings represent local, national, and international artists. Over more than 95 years, the collection has grown through direct purchases, private donations, and contributions from the Friends of the Museum Association. Currently, the most significant method of acquisition comes from the twelve Acquisition Awards granted annually by the Provincial Salon of Visual Arts of Entre Rios, which is organized by the museum. The institution also maintains the Gloria Montoya specialized art library, which holds over 2,000 titles.

Among its most significant pieces is the renowned permanent collection of works by Cesareo Bernaldo de Quiros. The museum also holds works by prominent Argentine artists such as Benito Quinquela Martin, Emilio Caraffa, and Juan Carlos Migliavacca. Five permanent rooms are dedicated to the heritage collection, including a hall specifically dedicated to plastic artists from the Province of Entre Rios. In addition to its permanent displays, the museum hosts temporary exhibitions in its four temporary rooms, such as a 1961 exhibition of Eduardo Seron's art and a 2013 temporary exhibition of works by the painter Joan Miro, organized in collaboration with the Cultural Office of the Embassy of Spain.

Since 1980, the museum has been permanently housed in the historic former residence of the Tezanos Pinto family, a building acquired by the Government of Entre Rios for this purpose. The architecture of the old house reflects the grandeur of the past, providing an elegant setting for the art within. The site carries profound historical weight, as the building was used as a clandestine detention and torture center during the Argentine dictatorship. In 2012, it was officially identified as a Site of Memory with a plaque placed on the premises to serve as a reminder of this period. The facility includes four rooms for temporary exhibitions, five rooms for permanent displays, and an auditorium.

The museum is located at Buenos Aires 355 in Parana, Argentina. It operates as a provincial institution under the Secretary of Culture of Entre Rios. Admission is free, making it an accessible cultural experience for the public. From October to March, operating hours are Tuesday through Friday from 8:00 to 13:00 and 15:00 to 20:00. The museum offers an annual program of guided tours, seminars, workshops, and film screenings oriented toward social and community inclusion.
